The substance having amino base such as albumin is mixed on the film 3 in advance for easy cross-link immobilization of enzyme. The enzyme is immobilized on the layer 3 by using glutaraldehyde, and the enzyme electrode 10 is made as the immobilization enzyme layer 4. The electrode 10 is immersed in the buffer solution 11 including the substance specifically reacted with enzyme and the concentration of substrate is measured by using the unit consisting of the reference electrode 7, salt bridge 8, pair electrode 9, potentiostat 6 and recorder 5. Thus, the analysis of substrate can be made by using only slight and expensive enzyme.
